Different dermal cell types have been reported to differ in their sensitivity to nano-sized TiO₂ . Kiss et al. exposed human keratinocytes (HaCaT), human dermal fibroblast cells, sebaceous gland cells (SZ95) and primary human melanocytes to 9 nm-sized TiO₂ particles at concentrations from 0.15 to 15 μg/cm² for up to 4 days. The particles were detected in the cytoplasm and perinuclear region in fibroblasts and melanocytes, but not in kerati-nocytes or sebaceous cells. The uptake was associated with an increase in the intracellular Ca²⁺ concentration. A dose- and time-dependent decrease in cell proliferation was evident in all cell types, whereas in fibroblasts an increase in cell death via apoptosis has also been observed. Anatase TiO₂ in 20–100 nm-sized form has been shown to be cytotoxic in mouse L929 fibroblasts. The decrease in cell viability was associated with an increase in the production of ROS and the depletion of glutathione. The particles were internalized and detected within lysosomes. In human keratinocytes exposed for 24 h to non-illuminated, 7 nm-sized anatase TiO_2, a cluster analysis of the gene expression revealed that genes involved in the “inflammatory response” and “cell adhesion”, but not those involved in “oxidative stress” and “apoptosis”, were up-regulated. The results suggest that non-illuminated TiO₂ particles have no significant impact on ROS-associated oxidative damage, but affect the cell-matrix adhesion in keratinocytes in extracellular matrix remodelling. In human keratinocytes, Kocbek et al. investigated the adverse effects of 25 nm-sized anatase TiO₂ (5 and 10 μg/ml) after 3 months of exposure and found no changes in the cell growth and morphology, mitochondrial function and cell cycle distribution. The only change was a larger number of nanotubular intracellular connections in TiO₂-exposed cells compared to non-exposed cells. Although the authors proposed that this change may indicate a cellular transformation, the significance of this finding is not clear. On the other hand, Dunford et al. studied the genotoxicity of UV-irradiated TiO₂ extracted from sunscreen lotions, and reported severe damage to plasmid and nuclear DNA in human fibroblasts. Manitol (antioxidant) prevented DNA damage, implying that the genotoxicity was mediated by ROS.

Titanium Dioxide is largely produced by the reduction of titanium tetrachloride, obtained in turn from chlorination of natural rutile, synthetic rutile derived from ilmenite or even slags rich in TiO2 produced by metallurgical treatment of ilmenite. TiO2 is also manufactured by treatment of ilmenite with sulfuric acid. Raw materials and the respective production processes employed in the manufacturing of Titanium Dioxide are listed below.

China’s government is working on its plan to eliminate outmoded capacities of TiO2 and strengthen environmental protection measurements. Small and middle-sized enterprises, with a production capacity under 50,000t/a, are the ones to suffer. They mostly get merged or need to withdraw from the market. Without their production capacity of about 500,000 tonnes yearly, the domestic output might shrink about 20%. This will enhance the power of listed companies in China immensely, which is one of the main reasons for the effort to go public.

Numerous studies have linked titanium dioxide to genotoxicity and cytotoxicity. Genotoxicity refers to a chemical’s potential to cause DNA damage, which can, in turn, lead to cancer. Cytotoxicity is a general term that refers to a characteristic of being harmful to cells.  

The morphology of vitaminB2@P25TiO₂NPs is coherent with the description of Degussa P25 typical population. Size distribution histograms were made from manual measures of the nanoparticles observed in SEM micrographs using ImageJ®. This data showed that more than 70% is anatase (between 20 and 60 nm) with a minor amount of rutile characteristic bars (between 80 and 100 nm) and a small amount of amorphous phase (<40 nm) [36]. Further analysis of the same sample areas with an EDS probe demonstrated the presence of organic material composed of C and O (Fig. 2). This material was found homogeneously distributed on the surface of the different shapes of P25TiO₂NP, not in the background, indicating a specific interaction that could be attributed to the functionalization of the P25TiO₂NPs with vitamin B2.

It adds a bright white color to coffee creamers, baked goods, chewing gums, hard-shell candies, puddings, frostings, dressings, and sauces. But the nanoparticles found in “food-grade” titanium dioxide may accumulate in the body and cause DNA damage—which is one way chemicals cause cancer and other health problems. 

Following the filtration, the precipitate is washed thoroughly to remove any impurities and excess reagents. This is a crucial step, as thorough washing ensures that no contaminants interfere with the final mass measurement. Once washed, the precipitate is dried in an oven at a controlled temperature to remove moisture, ensuring that the mass recorded reflects only the titanium content.

Other research suggests that E171 could cause harm; however, those research processes did not consider how people are typically exposed to E171. Research that adds E171 to drinking water, utilizes direct injections, or gives research animals E171 through a feeding apparatus is not replicating typical human exposure.
